If you are using the verbs попросить and спросить, after which verb can you use both кого and у кого?

Both verbs can be used with both "кого" and "у кого". However, the semantics may be different:
1. попросить кого-нибудь - ask somebody [to do something]
2. попросить у кого-нибудь - ask somebody [to give you something]
3. спросить кого-нибудь - ask somebody a question
4. спросить у кого-нибудь - the same as #3.

Please note that "попросить кого-нибудь" and "спросить кого-нибудь" may also be a [low-class] colloquial replacement of "попросить позвать кого-нибудь".

You probably refer to a phone call:
- Алло?
- Здравствуйте, попросите Ивана Николаевича/вы не могли бы попросить Ивана Николаевича (подойти к телефону - omitted)?

But it's not necessarily low-class. You may hear this at some office from a doctor or an official:
- Анна Ивановна (secretary's name), просите следующего (посетителя войти - omitted)
and it's very polite, nothing uneducated in it, probably quite the contrary.
