I work full-time, but would like to take classes on the side. Could I use the verb "подучиться" to describe this? If not, what word is best?

It's fine, but has a connotation of either some quick-and-dirty approach, or a refresher course for something that was once already learnt. If you mean that you'd like to study something seriously, then it's better to say "но мне хотелось бы поучиться/позаниматься помимо этого" or "в свободное время".
