Almost, but not quite. Literally, it means "at the end of the world". Could be an interesting and beautiful place, but a looong way from anywhere else. "Middle of nowhere" is, on the other hand, boring.

They sound a little differently but there is no difference in usage/meaning.
I think "на" reflects the English idiom better, though.

an idiom for "middle of nowhere" will be "у черта на куличках" (just don't ask who or what are кулички, no one knows that anyway).

The very idea that devil can have a "kulichi party" is very improbable, as is the "kulichi party" phrase. Kulich is not just a cake, it is, as you have mentioned, an exlusively Easter dish, i.e. made to celebrate resurrection of Christ. Now, why would devil celebrate Easter and Resurrection of Christ?

Not really; безлюдное место can be any place where there is no people at the moment, e.g. an underground parking of a huge building in the middle of a huge city, or a supermarket at midnight or anything like that. The place "in the middle of nowhere", on the other hand, can contain lots of people; this phrase is even applicable to a village or a small town, provided that they are geographically distant from any places of interest.
